Abstract:Objective: To investigate the effect of germacrone on the malignant biological behavior of colorectal cancer (CRC) cells based on the RhoA/ROCK signaling pathway. Methods: CRC SW480 cells were grouped into control group, L-germacrone group, M-germacrone group, H-germacrone group, Y-27632 group, and H-germacrone+LPA group. CCK-8 assay kit, flow cytometry, scratch assay, Transwell method, and Western blot were used to detect the proliferation activity, apoptosis, migration, invasion abilities, and RhoA/ROCK signaling pathway protein expression in SW480 cells. Results: Compared with the control group in terms of OD value, apoptosis rate, scratch healing rate, invasive cell number, RhoA, and ROCK protein expression, the L-gemone group, the M-gemone group, the H- Guimarone group, Y-27632 group cell OD values, scratch healing rate, invasive cell number, RhoA, ROCK protein expression was significantly decreased (P< 0.05) and apoptosis rate was significantly increased (P<0.05); Compared with the H-gemone group, the cell OD value (P<0.001), scratch healing rate (P<0.001), number of invasive cells (P<0.001), RhoA (P<0.001), and ROCK (P<0.001) protein expression were significantly higher (P<0.05) in the H-gimenone+LPA group, and the apoptosis rate (P<0.001) was significantly reduced (P<0.05). Conclusion: The effect of gimarone on the malignant biological behaviour of CRC cells may be achieved by inhibiting the RhoA/ROCK signalling pathway.
