Value Analysis of Maternal Serum Protein Expression and Quantitative Mass Spectrometry in Down Syndrome |

Abstract Objective: To screen maternal serum protein markers in Down's syndrome.To analyze the expression characteristics of maternal serum proteins in Down's syndrome.Objective to study the value of quantitative mass spectrometry in the determination of serum proteins in Down syndrome patients.Methods: 30 pregnant women who received Down's syndrome in the middle of the pregnancy from June 2013 to August 2017 were selected as the observation group.30 healthy pregnant women were used as the control group.Isotope labeled absolute and relative quantitative mass spectrometry (iTRAQ) was used to determine the relative expression of serum protein and serum protein.The ELISA method was used to verify the content of the candidate protein in the maternal serum.Results: There were 26 high credible proteins, including 18 upregulated proteins and 8 down regulated proteins, by iTRAQ quantitative mass spectrometry.The PANTHER software analysis showed that 26 proteins involved 5 functions, including protein binding, antioxidant activity, catalytic function, structure and molecular activity and transport function.Compared with the control group, the serum CP and SOD levels in the observation group were significantly higher than those in the control group, and the difference was statistically significant between P < 0.05. Conclusion: Quantitative mass spectrometry can identify the difference of serum protein markers in the mother body of Down syndrome, and the serum peroxidase -1 protein and serum ceruloprotein are expected to be a marker for the maternal serum protein of Down's syndrome.
