1409 LncRNA TUSC7 Inhibits Proliferation Migration and Invasion of Colorectal Cancer by Regulating the miR-182/TBX5 Axis

Objective: To investigate the inhibitory effects of long non-coding RNA tumor suppressor candidate 7 (LncRNA TUSC7) on the proliferation, migration, and invasion of colorectal cancer (CRC) via regulation of microRNA-182 (miR-182)/T-box5 (TBX5) axis.Methods: Quantitative real-time PCR was performed to detect the levels of LncRNA TUSC7, miR-182, and TBX5 in 90 pairs of CRC adenocarcinoma tissues and the corresponding adjacent normal tissues, and their correlations were analyzed in clinical samples, as well as the expressions of LncRNA TUSC7 in human CRC cell lines LoVo, SW1116, CaCO2, HCT116 and SW480 and normal human colon mucosal cell line NCM460. RNA fluorescence in situ hybridization was conducted to detect the subcellular localization of LncRNA TUSC7. SW480 cells were divided into 10 groups:TUSC7 knockdown (shTUSC7) group and knockdown control (shNC) group, TUSC7 overexpression (oeTUSC7) group and overexpression control (oeNC) group, miR-182 overexpression (miR-182 mimic) group and overexpression control (NC mimic) group, interference miR-182 (miR-182 inhibitor) group and interference control (NC inhibitor) group, simultaneous overexpression of TUSC7 and miR-182 (oeTUSC7+miR-182) group and simultaneous overexpression of TUSC7, miR-182, and TBX5 (oeTUSC7+miR-182 mimic+oeTBX5) group, transfected with shTUSC7, shNC, oeTUSC7, oeNC, miR-182 mimic, NC mimic, miR-182 inhibitor, NC inhibitor, oeTUSC7+miR-182 mimic, and oeTUSC7+miR-182 mimic+oeTBX5, respectively. The effects of LncRNA TUSC7, miR-182, and TBX5 on cell viability, proliferation, apoptosis, migration, and invasion of SW480 cells were determined by CCK8, clonal formation, flow cytometry, and Transwell assays. Dual luciferase reporter assay, RNA immunoprecipitation, RNA pull-down assay, and Western blot were performed to verify the relationships in LncRNA TUSC7, miR-182, and TBX5. Kaplan-Meier method was used to determine survival analysis. The nude mice were randomly divided into 4 groups (n=12):shTUSC7 group, shNC group, oeTUSC7 group, and oeNC group, which were inoculated subcutaneously with SW480 cells transfected with shTUSC7, shNC, oeTUSC7, and oeNC, respectively, in the right underarm. The effects of LncRNA TUSC7 on tumor growth of CRC and survival analysis were evaluated by xenograft model in vivo.Results: Compared to adjacent normal tissue, LncRNA TUSC7 (1.53±0.84 vs 6.97±3.97) and TBX5 (0.58±0.29 vs 3.25±1.51) were low-expressed, while miR-182 (0.54±0.33 vs 0.09±0.06) was high-expressed in CRC tissues (P<0.05). LncRNA TUSC7 was negatively correlated with miR-182 (R2=0.09077), but positively related to TBX5 (R2=0.08371). miR-182 were negatively associated with TBX5 (R2=0.06914;P<0.05). Compared with NCM460 (1.00±0.05) cells, LncRNA TUSC7 was significantly lower expressed in LoVo (0.51±0.08), SW1116 (0.41±0.10), CaCO2 (0.42±0.11), HCT-116 (0.53±0.10) and SW480 (0.35±0.07) cells (P<0.05). The expression was lowest in SW480 cells. LncRNA TUSC7 was located in the CRC cytoplasm. Compared with the shNC/oeNC group, silencing/overexpression of LncRNA TUSC7 promoted/inhibited the proliferation, migration, and invasion, while decreased/increased apoptosis of CRC cells. LncRNA TUSC7 directly targeted miR-182, and TBX5 was a direct target of miR-182. Mechanistically, LncRNA TUSC7 promoted the expression of TBX5 by targeting miR-182, thus inhibiting the proliferation, migration, and invasion, but promoting apoptosis of CRC cells (P<0.05). Consistent with the results in vitro, LncRNA TUSC7 could inhibit CRC growth in vivo.Conclusion: LncRNA TUSC7 was low expressed in CRC tissues and cells. LncRNA TUSC7 inhibited the proliferation, migration and invasion of CRC cells by regulating the miR-182/TBX5 axis.

1421 Mechanism of MiR-125a-5p Targeting FGFR1 and FGFR3 to Inhibit the Malignant Biological Behavior of Cervical Cancer Cells

Objective: To investigate the mechanism by which microRNA-125a-5p (miR-125a-5p) targets fibroblast growth factor receptor (FGFR) 1 and FGFR3 to inhibit the malignant biological behavior of cervical cancer (CC) cells.Methods: The qRT-PCR method was used to detect the expression of miR-125a-5p, FGFR1, and FGFR3 in CC tissue samples and adjacent tissue samples of 37 CC patients who underwent surgery in our hospital from June 2022 to June 2023. CC CaSKi cells were randomly separated into control group, NC-mimics group, miR-125a-5p-mimics group, miR-125a-5p mimics+pcDNA-NC group, miR-125a-5p mimics+pcDNA-FGFR1 group, and miR-125a-5p mimics+pcDNA-FGFR3 group. The expression of miR-125a-5p, FGFR1, and FGFR3 in each group was measured. MTT and plate cloning methods were applied to detect CaSKi cell proliferation. Transwell experiment was conducted to detect the migration and invasion of CaSKi cells. Flow cytometry was used to detect apoptosis of CaSKi cells. WB was applied to detect the expression of FGFR1 and FGFR3 proteins in CaSKi cells. A dual luciferase reporter gene experiment was applied to verify the relationship between miR-125a-5p and FGFR1, and between miR-125a-5p and FGFR3.Results: The expression of miR-125a-5p in CC tissue was lower than that in adjacent cancer tissue, while the expression of FGFR1 and FGFR3 was higher than that in adjacent cancer tissue (P<0.05). The apoptosis rate and miR-125a-5p expression in CaSKi cells in the miR-125a-5p-mimics group were higher than those in the control group and NC-mimics group, the EdU positive cell rate, OD490, migration number, invasion number, FGFR1 mRNA and protein, and FGFR3 mRNA and protein expression were lower than those in the control group and NC-mimics group (P<0.05). Compared with the miR-125a-5p-mimics group and the miR-125a-5p mimics+pcDNA-NC group, the apoptosis rate of the miR-125a-5p mimics+pcDNA-FGFR1 group decreased, the EdU positive cell rate, OD490, migration number, invasion number, FGFR1 mRNA and protein expression increased (P<0.05), the apoptosis rate of the miR-125a-5p mimics+pcDNA-FGFR3 group decreased, while the EdU positive cell rate, OD490, migration number, invasion number, FGFR3 mRNA and protein expression increased (P<0.05). MiR-125a-5p targeted negative regulation of FGFR1 and FGFR3.Conclusion: Overexpression of miR-125a-5p can inhibit the malignant biological behavior of CC cells, and its mechanism may be achieved by targeting FGFR1 and FGFR3.

1428 Effect of Paeoniflorin on Hydrogen Peroxide-Induced Oxidative Stress Damage in Skin Fibroblasts by Regulating the SIRT1/PGC-1α/Nrf2 Signaling Pathway

Objective: To investigate the effect of paeoniflorin (PF) on oxidative stress damage induced by hydrogen peroxide (H2O2) in skin fibroblasts (HSF) by regulating the silent information regulator 2 homologue 1 (SIRT1)/peroxisome proliferator activated receptor-gamma coactivator-1α (PGC-1α)/nuclear factor erythroid-derived 2-related factor 2 (Nrf2) signaling pathway.Methods: HSF cells were studied and separated into the Control group, H2O2 group, L-PF, M-PF, H-PF group, and PF+EX527 group. CCK-8 was applied to detect HSF cell proliferation. The β-galactosidase staining method was applied to detect HSF cell aging. Flow cytometry was used to detect apoptosis of HSF cells. ELISA method was used to detect the expression of ROS, SOD, GSH, and MDA in HSF cells. WB was used to detect the expressions of SIRT1, PGC-1α, and Nrf2 proteins in HSF cells.Results: The survival rate, SOD, GSH, SIRT1, PGC-1α, and Nrf2 expression of HSF cells in the H2O2 group were lower than in the control group, the proportion of aging cells, apoptosis rate, ROS, and MDA expression were higher than in the control group (P<0.05). Compared with the H2O2 group, the survival rate, SOD, GSH, SIRT1, PGC-1α, and Nrf2 expression were increased in the L-PF group, M-PF group, and H-PF group, the proportion of aging cells, apoptosis rate, ROS, and MDA expression were decreased (P<0.05). The survival rate, SOD, GSH, SIRT1, PGC-1α, and Nrf2 expression in the PF+EX527 group were lower than in the H-PF group, the proportion of aging cells, apoptosis rate, ROS, and MDA expression were higher than in the H-PF group (P<0.05).Conclusion: PF can inhibit H2O2-induced oxidative stress damage in HSF cells, and its mechanism may be achieved by activating the SIRT1/PGC-1α/Nrf2 signaling pathway.

1434 LncRNA MALAT1 Mediates Epigenetic Silencing of Systemic Lupus Erythematosus-Associated Anti-Inflammatory Genes through Recruitment of EZH2

Objective: To explore the correlation of long non-coding RNA (LncRNA) MALAT1 and methyltransferase EZH2 with SLE-associated anti-inflammatory factors and the molecular regulatory mechanism.Methods: Peripheral blood mononuclear cells (PBMC) were extracted from healthy controls (n=10) and SLE patients (n=10), and mRNA levels of LncRNA MALAT1, EZH2, and anti-inflammatory factors PTEN, Ets-1, and FOXO1 were detected by real-time fluorescence quantitative polymerase chain reaction (RT-qPCR). The correlation between LncRNA MALAT1, EZH2, and the expression of stress factors was verified respectively. After transfecting THP-1 cells with small interfering RNA (si-RNA), the expression of various anti-inflammatory factors was detected by RT-qPCR after LncRNA MALAT1 and EZH2 was knocked down, and the interaction between LncRNAMALAT1 and EZH2 was verified by RNA immunoprecipitation (RIP). After simultaneous knockdown of EZH2 and overexpression of LncRNAMALAT1, the 27th lysine (K27) trimethylation (me3) level of histone 3 (H3) was detected by Western blot (western blot).Results: Compared with healthy controls, PBMC LncRNAMALAT1 and EZH2 in peripheral blood of SLE patients were significantly overexpressed (P<0.05), anti-inflammatory factors PTEN, Ets-1, and FOXO1 were significantly decreased (P<0.05), and LncRNA MALAT1 and EZH2 were significantly negatively correlated with the anti-inflammatory expressions. The expressions of various anti-inflammatory factors were significantly increased after LncRNA MALAT1 and EZH2 were knocked down respectively (P<0.05). The RIP result shows that LncRNA MALAT1 and EZH2 combine. After overexpression of LncRNA MALAT1, the expression of anti-inflammatory factors was significantly decreased. Still, the expression of anti-inflammatory factors was increased and H3K27me3 was decreased considerably when EZH2 was knocked down and LncRNA MALAT1 was overexpressed at the same time (P<0.05).Conclusion: The expression of LncRNA MALAT1 and EZH2 is significantly negatively correlated with the expression of SLE- associated anti-inflammatory factors, and the molecular mechanism is related to LncRNA MALAT1 regulating the expression of H3K27me3 by recruiting EZH2.

1440 Effect of CircOGDH on Oxygen-Glucose Deprivation/Reoxygenation-Induced Microglial Cell Injury through Modulation of the miR-195-5p/PGAM5 Axis

Objective: To investigate the effect of circular RNA OGDH (CircOGDH) on microglia (MG) cell damage induced by oxygen-glucose deprivation/reoxygenation (OGD/R) by modulating the miR-195-5p/phosphoglycerate mutase 5 (PGAM5) axis.Methods: HMC3 cells cultured in vitro were separated into:NC group (normal culture), OGD/R, si-NC group, si-CircOGDH group, mimic NC group, miR-195-5p mimic group, si-CircOGDH+inhibitor NC group, and si-CircOGDH+miR-195-5p inhibitor group. Except for the NC group, the remaining groups constructed OGD/R models before transfecting the corresponding substances. QRT-PCR method was applied to determine the expression levels of CircOGDH, miR-195-5p, and PGAM5 mRNA. The levels of cell proliferation, apoptosis, reactive oxygen species (ROS), and inflammatory factors were measured in each group. Western blot was applied to detect the expression of Cleaved caspase-3 and Bax. Dual luciferase gene experiment was used to detect the targeting relationship between CircOGDH and miR-195-5p, and between PGAM5 and miR-195-5p.Results: Compared with the NC group, the expression of miR-195-5p and OD450 value were reduced in the OGD/R group, the mRNA expression of CircOGDH and PGAM5, apoptosis rate, ROS, and inflammatory cytokine levels, and the expression of Cleaved caspase-3 and Bax proteins were all elevated (P<0.05). Compared with the OGD/R group and the si-NC group, the expression of miR-195-5p and OD450 value were elevated in the si-CircOGDH group, the mRNA expression of CircOGDH and PGAM5, apoptosis rate, ROS and inflammatory cytokine levels, and the expression of Cleaved caspase-3 and Bax proteins were all reduced (P<0.05). Compared with the OGD/R group and the mimic NC group, the miR-195-5p mimic group showed no significant difference in CircOGDH (P>0.05), while the expression of miR-195-5p and OD450 values were increased, the expression of PGAM5 mRNA, apoptosis rate, ROS and inflammatory cytokine levels, and the expression of Cleaved caspase-3 and Bax protein were all reduced (P<0.05). Compared with the si-CircOGDH+inhibitor NC group, the CircOGDH of the si-CircOGDH+miR-195-5p inhibitor group was not significantly different (P>0.05), the expression of miR-195-5p and OD450 values were decreased, the expression of PGAM5 mRNA, apoptosis rate, ROS and inflammatory cytokine levels, and the expression of Cleaved caspase-3 and Bax protein were all elevated (P<0.05). A dual luciferase gene experiment showed that miR-195-5p had a targeting relationship with CircOGDH and PGAM5.Conclusion: Silencing CircOGDH can promote the proliferation of HMC3 cells induced by OGD/R, inhibit their inflammatory response and apoptosis, and may be related to the regulation of miR-195-5p/PGAM5 axis.

1446 Effect of Emodin on PI3K/Akt/Mtor Signaling Pathway in Hippocampus of Schizophrenic Rats

Objective: To investigate the impact of emodin on neurological injury and cognitive function in rats with schizophrenia (SP) by regulating the phosphatidylinositol3-kinase (PI3K)/protein kinase B (Akt)/mammalian target of rapamycin (mTOR) signaling pathway.Methods: An SP rat model was established by injecting dexamethasone (MK-801), and normal rats were used as the control group. SP rats were randomly included in the SP group, different doses (20, 40, 60mg/kg) of emodin groups, and 60mg/kg of emodin+LY294002 group. Stereotypic behavior and ataxia scoring criteria were applied to evaluate rat behavioral function. Morris water maze experiment was applied to evaluate cognitive function in rats. Reagent kits were applied to detect acetylcholinesterase (AchE) and acetylcholine (Ach) activities in brain tissue. HE was applied to detect pathological changes in hippocampal tissue. QRT-PCR was applied to detect the expression of brain-derived neurotrophic factor (BDNF) mRNA and calcium-dependent protein kinase (CaMK II) mRNA in brain tissue. Western blot was applied to detect the expression of PI3K/Akt/mTOR pathway-related proteins.Results: Compared with the control group, the number of platform crossings, Ach activity, the expression of BDNF mRNA, CaMK Ⅱ mRNA, the expression of p-PI3K/PI3K, p-Akt/Akt, and p-mTOR/mTOR were greatly reduced in the SP group, the stereotypes and ataxia scores, average avoidance latency, and AchE activity were greatly increased (P<0.05). However, after intervention with different doses of emodin, the number of platform crossings, Ach activity, the expression of BDNF mRNA, CaMK Ⅱ mRNA, the expression of p-PI3K/PI3K, p-Akt/Akt, and p-mTOR/mTOR were greatly increased, the stereotypes and ataxia scores, average avoidance latency, and AchE activity were greatly reduced, with differences between groups (P<0.05). The number of platform crossings, Ach activity, the expression of BDNF mRNA, CaMK Ⅱ mRNA, the expression of p-PI3K/PI3K, p-Akt/Akt, and p-mTOR/mTOR in 60mg/kg emodin + LY294002 group were greatly lower than 60mg/kg emodin group, the stereotypes and ataxia scores, average avoidance latency, and AchE activity were greatly increased (P<0.05).Conclusion: Emodin can improve the cognitive ability and neuroprotective function of SP rats by regulating PI3K/Akt/mTOR signaling pathway.

1452 Plantamajoside Attenuates LPS-Induced Inflammatory Response of Human Gingival Fibroblasts by Inhibiting the NF-κB/IL-6 Signaling Pathway

Objective: To explore the effect of plantamajoside (PMS) on the inflammatory response of human gingival fibroblasts induced by lipopolysaccharide (LPS) by inhibiting nuclear factor-κB (NF-κB)/interleukin-6 (IL-6) signaling pathway.Methods: The cultured human gingival fibroblasts (HGnF) were randomly divided into control group, model group (10mg/L LPS), PMS low and high-dose groups (50, 100μg/mL PMS+10mg/L LPS), activator group (10μM BAY11-7085+10mg/L LPS), and PMS high-dose + activator group (10μM BAY11-7085+100μg/mL+10mg/L LPS). CCK-8 method was used to detect HGnF cell viability, ELISA method was used to detect the contents of interleukin 1β (IL-1β), interleukin 6 (IL-6), and interleukin 18 (IL-18) in the supernatant of HGnF cells. Western blot method was used to detect the expression levels of inducible nitric oxide synthase (iNOS), IL-1β and NF-κB/IL-6 signaling pathway-related proteins.Results: Low and high-doses PMS could increase the cell viability of HGnF induced by LPS, decrease the contents of IL-1β, IL-6, IL-18 in supernatant, and the expression levels of IL-1β, iNOS, p-NF-κB/NF-κB, and IL-6 in cells (P<0.05). The activator BAY11-7085 reversed the effects of high-dose PMS on the above indicators in LPS-induced HGnF cells (P<0.05). Conclusion: PMS may improve the inflammation of HGnF cells induced by LPS by inhibiting the NF-κB/IL-6 signaling pathway.

1456 Improvement Effect of Baicalin on Depression-Like Behavior in Depression Rats by Regulating the AMPK/mTOR Signaling Pathway

Objective: To investigate the improvement effect and mechanism of Wogonin (wog) on depression-like behavior in depression rats by regulating the AMP-activated protein kinase (AMPK)/mammalian target of rapamycin (mTOR) signaling pathway.Methods: A depression rat model was established, and the experiment was separated into control group, model group, low, medium, and high dose wogonin groups (Wog-L, Wog-M, Wog-H groups, 7mg·kg-1·d-1, 14mg·kg-1·d-1, 28mg·kg-1·d-1 Wog), and high-dose wogonin+AMPK inhibitor Compound C group (Wog-H+Compound C group, 28mg·kg-1·d-1 Wog+0.14mg·kg-1·d-1 Compound C). Sucrose preference, open field experiment, and forced swimming experiments were applied to observe depression-like behavior in rats; ELISA was applied to measure DA and 5-HT levels; transmission electron microscopy was applied to observe the ultrastructure and autophagy changes of hippocampal tissue; Nissl staining was applied to observe the morphological changes of neurons in the hippocampal CA1 region; immunohistochemistry was applied to detect the BDNF expression; Western blot was applied to detect the expression of AMPK, p-AMPK, mTOR, p-mTOR, P62, and Beclin-1. Results: Compared with the control group, the model group had sparse neuronal arrangement, accompanied by neuronal loss and inflammatory cell infiltration, widened cell gaps, reduced Nissl granules, lighter staining, severe swelling of mitochondria and neuronal cytoplasm, blurred mitochondrial boundaries, expansion of endoplasmic reticulum and Golgi apparatus, and reduced number of autophagic vesicles, the sugar preference, standing time, movement distance, the levels of DA and 5-HT, BDNF optical density values, and the expression of Beclin-1 and p-AMPK/AMPK were obviously reduced, the resting time and the expression of P62 and p-mTOR/mTOR were obviously increased (P<0.05). Compared with the model group, the morphology of neurons in WAG-L, WAG-M, and WAG-H groups was significantly improved, and the sugar water preference, standing time, moving distance, DA and 5-HT levels, BDNF optical density and Beclin-1 and p-AMPK/AMPK expression were significantly increased, the rest time and expression of P62 and P-MTOR /mTOR were significantly decreased (P<0.05). Compound C reversed the improvement effect of baicalin on depression in rats (P<0.05).Conclusion: Wogonin can improve depression-like behavior in rats with depression, and its mechanism may be related to regulating the AMPK/mTOR signaling pathway and enhancing autophagy.

1462 Correlation of Plasma Levels of RANTES and MST1 with Coronary Plaque Burden in Patients with Unstable Angina

Objective: To investigate the correlation between plasma levels of regulated upon activation, normal T-cell expressed and secreted (RANTES) and mammalian sterile 20-like kinase 1 (MST1) and coronary plaque burden in patients with unstable angina (UA).Methods: Hospitalized patients diagnosed with UA who underwent coronary angiography in the Department of Cardiology at the Affiliated Hospital of Chengde Medical College from January 2018 to January 2021 were selected. Enzyme-linked immunosorbent assay (ELISA) was used to detect plasma levels of RANTES and MST1 at admission. The extent of coronary plaque burden was quantified using the SYNTAX score and Gensini score. Patients were divided into low SYNTAX score group (≤22) and medium/high SYNTAX score group (>22). Additionally, patients were divided into low RANTES group, medium RANTES group, and high RANTES group, as well as low MST1 group, medium MST1 group, and high MST1 group based on tertiles of RANTES and MST1 levels.Results: A total of 321 cases UA patients were included in the study. Plasma levels of RANTES and MST1 were significantly higher in the medium/high SYNTAX score group compared to the low SYNTAX score group (P<0.05). The SYNTAX score was higher in the high RANTES group (18.91±6.57) than in the medium RANTES group (13.69±3.05) and the low RANTES group (10.66±6.12) (P<0.05). The SYNTAX score was higher in the high MST1 group (15.75±6.13) than in the medium MST1 group (14.43±7.51) and the low MST1 group (13.06±5.28) (P<0.05). Multivariate logistic regression analysis indicated that the highest tertiles of RANTES (OR=2.67, 95%CI 1.17~6.09, P<0.05) and MST1 (OR=5.274, 95%CI 1.769~15.722, P<0.05) were independent risk factors for medium/high SYNTAX scores. The area under the receiver operating characteristic (ROC) curve (AUC) for predicting medium/high SYNTAX scores was 0.69 (95%CI0.586~0.795, P<0.05) for RANTES,0.625 (95%CI0.553~0.696, P<0.05) for MST1, and 0.786 (95%CI0.705~0.866, P<0.001) for the combination of RANTES and MST1.Conclusion: Elevated plasma levels of RANTES and MST1 are independent risk factors for higher coronary plaque burden in patients with UA.

Objective: To investigate the effect of edaravone dexborneol (EDDE) on neurological function in rats with cerebral ischemic stroke (CIS), and to preliminarily explore the possible mechanism of BDNF (brain-derived neurotrophic factor) -AKT-CREB (cAMP response element binding protein) signaling pathway.Methods: Rats were randomly divided into five groups:sham operation group (sham group), CIS model group (CIS group), EDDE low dose group (EDDE-L group), EDDE high dose group (EDDE-H group), and nimodipine group; neural function was assessed by mNSS scoring method, and the area of cerebral infarction was detected by 2,3,5-triphenyltetrazolium chloride (TTC) staining in the rat, and the area of ni The pathological damage of rat brain tissue was observed, and the interleukin-1β (IL-1β), interleukin-6 (IL-6), superoxide dismutase (SOD), malondialdehyde (MDA) content, BDNF mRNA, AKT mRNA, CREB mRNA, and BDNF, AKT, CREB protein expression of brain tissue were also detected.Results: Compared with the sham group, the neurological function score, cerebral infarction area, apoptosis rate and the contents of IL-1β, IL-6, and MDA in the CIS group were significantly increased, while the expressions of SOD, BDNF mRNA, AKT mRNACREB mRNA, BDNF protein, AKT protein, CREB protein were significantly decreased (P<0.05). At the same time the number of cells in the cerebral cortex decreases, vacuoles are more numerous and nuclei appear to be consolidated. Compared with the CIS group, the neurological function scores, cerebral infarct area, apoptosis rate, and the contents of IL-1β, IL-6, and MDA were significantly reduced in the EDDE-L, EDDE-H, and nimodipine groups, whereas the contents of SOD, the BDNF mRNA, AKT mRNA, CREB mRNA, and the expression of BDNF protein, AKT protein, and CREB protein were all significantly increased ( P<0.05), and the cellular structure of the cerebral cortex was also relatively intact; among them, the effect was more obvious in the EDDE-H and nimodipine groups (P<0.05).Conclusion: EDDE may reduce the area of cerebral infarction and improve the neurological impairment in CIS rats by activating the BDNF-AKT-CREB pathway, inhibiting the expression of inflammatory factors, and reducing the level of oxidative stress.

1473 Effects of Theaflavin on Proliferation Apoptosis and Autophagy of A549 Cell by Regulating the PI3K/Akt/mTOR Signaling Pathway

Objective: To investigate the effects of theaflavin on A549 cell proliferation, apoptosis, and autophagy by regulating the phosphatidylinositol 3-kinase (PI3K)/protein kinase B (AKT)/mammalian target of rapamycin (mTOR) signaling pathway.Methods: The proliferation rate of A549 cells was measured after intervention with 0, 2.5, 5, 10, 20, 40, 80, and 160 μmol/L theaflavin, and the appropriate concentration of action was screened. A549 cells were separated into control group, low theaflavin group (20 μmol/L theaflavin), medium theaflavin group (40 μmol/L theaflavin), high theaflavin group (80 μmol/L theaflavin), and high theaflavin+740Y-P group (80 μmol/L theaflavin+30 μmol/L PI3K/Akt/mTOR signaling pathway activator 740Y-P). CCK-8 method and plate colony formation experiment were applied to determine the proliferation of A549 cells. Flow cytometry and MDC methods were applied to determine apoptosis and autophagic vacuole formation in A549 cells, respectively. Western blot was applied to determine the expression of PI3K/Akt/mTOR signaling pathway proteins and autophagy proteins in A549 cells.Results: Compared with 0 μmol/L of theaflavin, 2.5, 5, 10, 20, 40, 80, and 160 μmol/L of theaflavin can inhibit the proliferation of A549 cells. The IC50 value of theaflavin on A549 cells was calculated to be 36.72 μmol/L. Compared with the control group, the proliferation rate, colony formation rate, p-PI3K/PI3K, p-Akt/Akt, and p-mTOR/mTOR of A549 cells in the low theaflavin group, medium theaflavin group, and high theaflavin group all reduced, the apoptosis rate, relative content of autophagic vacuoles, LC3II/LC3I, and Beclin-1 protein expression all increased, and all showed dose-dependent changes in theaflavin (P<0.05). Compared with the high theaflavin group, the proliferation rate, colony formation rate, p-PI3K/PI3K, p-Akt/Akt, and p-mTOR/mTOR of A549 cells in the high theaflavin+740Y-P group all increased, the apoptosis rate, relative content of autophagic vacuoles, LC3II/LC3I, and Beclin-1 protein expression all reduced (P<0.05).Conclusion: Theaflavin can induce protective autophagy by inhibiting the activation of the PI3K/Akt/mTOR signaling pathway, thereby promoting apoptosis and inhibiting the proliferation of lung cancer cells.

1478 BMSCs Exosomes-Derived miR-181 Promotes Osteogenic Differentiation in Mice with Femoral Fracture by Targeting ERK5

Objective: To investigate the regulatory effects of BMSCs-derived exosomes (BMSCs-Exos) on osteogenic differentiation in vitro, to elucidate the mechanism by which miR-181 targets ERK5 to mediate this regulation, and to study the impact of BMSCs-Exos and overexpressed miR-181 on femoral fracture healing in mice.Methods: BMSCs-Exos were characterized using dynamic light scattering (DLS) and Western blot techniques. The expression levels of miR-181 and ERK5 were assessed via qRT-PCR and Western blot. Dual-luciferase reporter assays were performed to validate the targeting relationship between miR-181 and ERK5. Protein expression and ALP activity were analyzed using Western blot and ELISA. In a mouse femoral fracture model, miR-181 mimic or BMSCs-Exos were injected, and fracture healing was evaluated by measuring callus volume (CV), trabecular bone volume fraction (BV/TV), and relevant protein levels.Results: The particle size of BMSCs-Exos ranged from 50 to 150 nm, with a Zeta potential of -25.69±2.88 mV. Surface markers CD9, CD63, and CD81 were prominently expressed. In the BMSCs-Exos group, miR-181 expression was upregulated (P<0.05), while ERK5 expression was downregulated (P<0.05). The miR-181 mimic increased miR-181 expression and decreased ERK5 protein levels (P<0.05). BMSCs-Exos delivering miR-181 promoted osteogenic differentiation of BMSCs in vitro by targeting ERK5 (P<0.05). Both BMSCs-Exos and overexpressed miR-181 enhanced femoral fracture healing in mice (P<0.05).Conclusion: BMSCs-Exos facilitate osteogenic differentiation of BMSCs in vitro by transporting miR-181 and targeting ERK5, and they also aid in femoral fracture healing in mice.

1483 Study on the Relationship between Neutrophils/Lymphocytes CD4+/CD8+ and the Efficacy and OS of PD-1 Inhibitor Immunotherapy for Lung Cancer

Objective: To observe the changes in neutrophil/lymphocyte ratio (NLR) and CD4+/CD8+ in patients with lung cancer after treatment with programmed death receptor-1 (PD-1) inhibitor, and to analyze the correlation between changes of two indicators and efficacy and overall survival time (OS).Methods: A total of 94 patients with pulmonary cancer were included in the hospital from December 2019 to March 2022. The changes of NLR and CD4+/CD8+ were observed among patients with different efficacies after PD-1 treatment, and the association with different efficacies after immunotherapy and OS after 2 years of follow-up was analyzed.Results: There were statistically significant differences in NLR and CD4+/CD8+ among patients with different efficacies after PD-1 inhibitor treatment (P<0.05). The NLR and CD4+/CD8+ in the CR group and PR group were statistically different from those in the SD group and CD group (P<0.05), and there was a statistical significance in NLR between the SD group and PD group (P<0.05). Univariate analysis showed that the pathological type, TNM staging, differentiation degree, age, NLR and CD4+/CD8+ were statistically significant between the effective group and the ineffective group after immunotherapy (P<0.05). Logistic regression model found that small cell type, TNM staging (stage IV), differentiation degree (low differentiation), older age, and high NLR were risk factors for poor efficacy of immunotherapy (P<0.05), while high CD4+/CD8+ was a protective factor for poor efficacy of immunotherapy (P<0.05). ROC curve showed that the areas under the curves (AUCs) of NLR and CD4+/CD8+ in evaluating the efficacy were 0.785 and 0.777 respectively, and the AUC of NLR combined with CD4+/CD8+ was 0.871, which was significantly higher than that of single index (P<0.05). Spearman correlation analysis suggested that NLR was negatively correlated with OS (r=-0.341, P<0.05), and CD4+/CD8+ was positively correlated with OS (r=0.323, P<0.05).Conclusion: The NLR is lower while the CD4+/CD8+ is higher among patients with pulmonary cancer who are with effective PD-1 inhibitor treatment, and the two indexes have certain evaluated values on the efficacy of immunotherapy and also have a certain association with OS.

1488 Effects of LncRNA LBX2-AS1 on the Proliferation Migration and Invasion of Gastric Cancer Cells by Regulating the miR-873-5p/G6PD Axis

Objective: To investigate the effects of long non-coding RNA (LncRNA) ladybird homeobox 2 antisense RNA1 (LBX2-AS1) on the proliferation, migration, and invasion of gastric cancer (GC) cells by regulating the micro RNA-873-5p (miR-873-5p)/glucose-6-phosphate dehydrogenase (G6PD) axis.Methods: SGC7901 cells were randomly separated into control group, sh-NC group, sh-LBX2-AS1 group, sh-LBX2-AS1+inhibitor NC group, and sh-LBX2-AS1+miR-873-5p inhibitor group. qRT-PCR method was applied to detect the expression of LncRNA LBX2-AS1, miR-873-5p, and G6PD. MTT method and plate cloning experiment were applied to detect the proliferation of SGC7901 cells. Scratch experiment was applied to detect the migration of SGC7901 cells. Transwell experiment was applied to detect the invasion of SGC7901 cells. Western blot was applied to detect the expression of MMP-2, PCNA, MMP-9, and G6PD proteins in SGC7901 cells. Luciferase reporter gene assay was applied to detect the interaction between LncRNA LBX2-AS1 and miR-873-5p, and between miR-873-5p and G6PD. Mouse tumor bearing experiment was applied to verify the effects of knocking out LncRNALBX2-AS1 on CC tumor growth and G6PD expression.Results: The A490 value, clone number, scratch healing rate, invasion number, LncRNA LBX2-AS1, G6PD mRNA and PCNA, MMP-2, MMP-9 protein expression in SGC7901 cells in sh-LBX2-AS1 group were lower than those in sh-NC group and control group, while miR-873-5p expression was higher than that in sh-NC group and control group (P<0.05). Compared with the sh-LBX2-AS1 group and the sh-LBX2-AS1+inhibitor NC group, the sh-LBX2-AS+miR-873-5p inhibitor group showed a decrease in miR-873-5p expression, and an increase in A490 value, clone number, scratch healing rate, invasion number, G6PD mRNA and PCNA, MMP-2, MMP-9 protein expression (P<0.05). LncRNA LBX2-AS1 targeted the negative regulation of miR-873-5p, and miR-873-5p targeted the negative regulation of G6PD. The quality, volume, Ki-67 positive rate, and G6PD positive rate of transplanted tumors in the experimental group were lower than those in the control group (P<0.05).Conclusion: Knocking out LncRNA LBX2-AS1 may inhibit the proliferation, migration, and invasion of GC cells, and its mechanism may be achieved by regulating the miR-873-5p/G6PD axis.

Objective: To compare the effect and safety of ultrasound-guided erector spinal muscle plane block anesthesia (ESPB) with epidural anesthesia during painless labor analgesia.Methods: Retrospectively, 80 women admitted from June 2022 to June 2023 were divided into group B (epidural anesthesia, 40); comparing the first stage and second stage, neonatal Apgar score (at birth and 5 min), maternal adverse reactions (headache, nausea, vomiting, pruritus, fever), and numerical rating scale (NRS) score in different periods (30 min before analgesia, 10 cm at cervical dilatation, and at delivery).Results: Stage of labor: comparing the first stage of labor in groups A and B, the difference was not significant (P>0.05); the second stage of labor in group A was shorter than group B, and the difference was significant (P<0.05). Apgar score: Groups A and B at 5 min Apgar, the difference was not significant (P>0.05). Adverse reactions: comparing the incidence of headache, nausea, vomiting, itching, and fever in groups A and B, the difference was not significant (P>0.05). NRS score: comparing the NRS score before analgesia, the difference between the two groups was not significant (P>0.05); the NRS score of 30 min analgesia, 10 cm cervical dilation was significantly lower than the difference (P<0.05); the NRS score at 30 min analgesia and 10 cm cervical dilation was lower than group B, and the difference was significant (P<0.05).Conclusion: In painless labor and analgesia, both ultrasus-guided ESPB anesthesia and epidural anesthesia have good effects and high safety, and ultrasus-guided ESPB anesthesia can further shorten the second stage of labor and have better analgesic effect.

1499 Application of Embryo Morphokinetic Parameters in the Construction of Embryonic Prediction Model

Objective: To establish a prediction model for euploidy based on embryo morphokinetic parameters, so as to provide a reference for evaluating embryonic developmental potential in assisted reproductive technology.Methods: A retrospective analysis was performed on the clinical data of patients who underwent pre-implantation genetic testing between December 2020 and June 2023. A total of 1383 available blastocysts from 364 cycles were included. According to euploidy, the blastocysts were divided into the euploidy group (442 blastocysts) and the non-euploidy group (941 blastocysts). The morphokinetic parameters of the two groups were compared, and multivariate logistic regression analysis was performed. A prediction model for euploidy was constructed. The ROC curve was used to analyze the sensitivity and specificity of the prediction model. Meanwhile, morphokinetic parameters of 658 available blastocysts from 215 cycles included from July 2023 to April 2024 were used for clinical validation. The consistency between the prediction model and clinical practice was analyzed.Results: t4, t5, s2, time of starting blastulation (tSB), time of hatching blastocyst (tHB), and tHB-tSB in the euploidy group were shorter than those in the non-euploidy group (P<0.05). Logistic regression analysis found that t4, t5, s2, tSB, tHB, and tHB-tSB were influencing factors of blastocyst euploidy (P<0.05). A prediction model for euploidy was constructed as follows:P==ex/(1+ex), X=-3.299-0.026×t4-0.018×t5-0.264×s2-0.009×tSB-0.007×tHB-0.153×tHB-tSB. Hosmer-Lemeshow goodness of fit test showed that the prediction model had good goodness of fit (χ2=8.700, P=0.368). The area under the ROC curve of the prediction model was 0.871 (95%CI=0.852~0.889, P<0.001). Its sensitivity and specificity were 75.86% and 82.11%. Clinical validation indicated that the accuracy of the prediction model was 79.94%. The consistency between the prediction model and clinical practice was moderate (Kappa=0.561).Conclusion: Embryo morphokinetic parameters (t4, t5, s2, tSB, tHB and tHB-tSB) are related to the formation of euploidy. The prediction model constructed based on embryo morphokinetic parameters in this study is of high value in predicting embryo euploidy and can provide a reference for evaluating embryonic developmental potential.

1505 Association of Placental Growth Factor Levels with Fetal Cardiac Monitoring and Intrauterine Fetal Growth Restriction in Pregnant Women with Pre-Eclampsia

Objective: To analyze the association between placental growth factor level and fetal heart rate monitoring and fetal intrauterine growth restriction in pregnant women with preeclampsia. Methods: A total of 131 pregnant women with preeclampsia diagnosed in the hospital were selected from June 2020 to June 2023 as the research subjects. According to whether they were complicated with fetal intrauterine growth restriction, they were divided into preeclampsia without fetal growth restriction group (n=70) and preeclampsia with fetal growth restriction group (n=61). All study subjects were tested for placental growth factor level, and fetal heart rate monitoring was performed at more than 28 weeks of gestation. The receiver operating characteristic (ROC) curve was used to analyze the evaluated value of the placental growth factor on fetal intrauterine growth restriction in pregnant women with preeclampsia.Results: The level of placental growth factor in preeclampsia with fetal growth restriction group was lower than that in preeclampsia without fetal growth restriction group (P<0.05). The abnormal rate of fetal heart rate monitoring was higher in preeclampsia with fetal growth restriction group than that in preeclampsia without fetal growth restriction group (P<0.05). ROC curve analysis showed that the area under the curve (AUC), sensitivity, and specificity of placental growth factor in evaluating fetal intrauterine growth restriction in pregnant women with preeclampsia were 0.797, 91.80%, and 57.14%.Conclusion: The level of placental growth factor is closely related to fetal heart rate monitoring and fetal intrauterine growth restriction in pregnant women with preeclampsia. Placental growth factor detection and fetal heart rate monitoring can provide a reference for the evaluation of fetal intrauterine growth restriction in pregnant women with preeclampsia.

Objective: To analyze the predictive value of blood urea nitrogen (BUN), serum creatinine (SCr), and cystatin C (CysC) for acute kidney injury (AKI) in asphyxiated full-term neonates.Methods: The clinical data of 85 asphyxiated full-term neonates born in the hospital from January 2020 to June 2023 were collected retrospectively. According to the presence or absence of AKI, neonates selected were divided into the AKI group (n=30) and the non-AKI group (n=55). The clinical data, BUN, SCr, and CysC of the two groups were compared. Multivariate logistic regression analysis was performed to identify the risk factors for AKI in asphyxiated full-term neonates. The receiver operating characteristic (ROC) curves were used to analyze the predictive value of BUN, SCr, and CysC for AKI in asphyxiated full-term neonates.Results: There was no statistically significant difference in gender, gestational age, birth weight, delivery mode, or number of births between the two groups (P>0.05). The proportion of severe asphyxia and the levels of BUN, SCr, and CysC in the AKI group were higher than those in the non-AKI group (P<0.05). Logistic regression analysis found that severe asphyxia and abnormally elevated levels of BUN, SCr and CysC were independent factors in predicting the development of AKI in term neonates with asphyxia (P<0.05). ROC curves indicated that BUN, SCr, and CysC had certain predictive values for AKI in asphyxiated full-term neonates. Among them, the area under the curve (AUC) and sensitivity of CysC were higher than those of BUN and SCr (P<0.05).Conclusion: BUN, SCr, and CysC level tests are of relevance in guiding the development of AKI in full-term asphyxiated neonates, and serum CysC has the best guiding value.

1514 Relationship between Expressions of Serum miR-22 and miR-98-5p and Efficacy of Three-Dimensional Conformal Radiotherapy for Non-Small Cell Lung Cancer

Objective: To observe the differences in the expression levels of microRNA (miR)-22 and miR-98-5p in patients with non-small cell lung cancer (NSCLC) and different efficacies after three-dimensional conformal radiotherapy, and to analyze the association between expression levels and efficacy.Methods: A total of 104 patients with NSCLC who were admitted to the hospital from January 2020 to January 2024 were enrolled. All patients received three-dimensional conformal radiotherapy. After radiotherapy completion, the expression levels of miR-22 and miR-98-5p were determined by real-time fluorescence quantitative PCR. One-way analysis of variance was used to analyze the differences in the expression levels of miR-22 and miR-98-5p in patients with different efficacies. Spearman correlation analysis was applied to investigate the correlation with radiotherapy efficacy. The logistic multivariate regression model was adopted to analyze the related factors affecting the efficacy of radiotherapy. The receiver operating characteristic (ROC) curve was drawn to explore the evaluated efficiency of miR-22 and miR-98-5p expression levels on the effectiveness of radiotherapy.Results: There were statistically significant differences in the expression levels of miR-22 and miR-98-5p among patients with different efficacies (P<0.05). There was a statistical significance in the expression level of miR-22 between the SD group and the PD group (P<0.05). The expression levels of miR-22 and miR-98-5p revealed statistical differences in the CR group and PR group compared with those in the SD group and PD group (P<0.05). There were statistical differences in the expression levels of miR-22 and miR-98-5p between CR group and PR group (P<0.05). Univariate analysis showed that different pathological types, TNM stages, differentiation degrees, and expression levels of miR-98-5p and miR-22 were related to the efficacy of radiotherapy (P<0.05). Logistic regression model analysis revealed that TNM staging (OR=3.360, 95%CI:1.229-9.184), differentiation degree (OR=2.779, 95%CI:1.066-7.246), miR-98-5p expression level (OR=2.085, 95%CI:1.147-3.792), and miR-22 expression level (OR=0.590, 95%CI:0.368-0.946) were the influencing factors for evaluating the efficacy of radiotherapy (P<0.05). ROC curve suggested that the areas under the curves (AUCs) of miR-98-5p and miR-22 in evaluating the effectiveness of radiotherapy were 0.798 and 0.800, respectively, the sensitivities were 0.589 and 0.630, and the specificities were 0.871 and 0.839 respectively. The AUC of miR-98-5p+miR-22 was 0.895, significantly higher than that of a single indicator (P<0.05), and the sensitivity and specificity were 0.726 and 0.935.Conclusion: There are some differences in the expression levels of serum miR-98-5p and miR-22 among patients with different efficacies after three-dimensional conformal radiotherapy for NSCLC, and the expression levels are significantly related to the effectiveness of radiotherapy and have certain evaluated value on efficacy.

1519 Construction of a Nomogram Prediction Model for Failure of Extubation in Severe Pneumonia Based on Diaphragmatic Ultrasound Cardiac Parameters and Blood Gas Indices

Objective: To explore the value of combined phrenic ultrasonography, cardiac parameters, and blood gas indexes in predicting the failure of offline extubation of severe pneumonia, and to construct a nomogram prediction model for early clinical intervention.Methods: A total of 210 patients with severe pneumonia in the Affiliated Hospital of Guizhou Medical University from March 2021 to December 2023 were selected as the study subjects, and the study subjects were randomly divided into a training set (70%, 147 cases) and a verification set (30%, 63 cases). The general information of the patients was analyzed. The lasso-Logistic regression equation was used to screen the predictors of offline extubation failure for severe pneumonia, and a nomogram prediction model was constructed. The receiver operating characteristic curve (ROC), decision curve (DCA), and calibration curve were used to analyze the efficacy of the model.Results: In the training set and the validation set, the differences in age, mechanical ventilation time, APACHE Ⅱ score, ICU stay, CRP/ALB, cardiac parameters, diaphragm ultrasound parameters, PaO2, PaCO2, P/F, PA-aO-2, and history of underlying cardiopulmonary disease were statistically significant when comparing the case group (failed extubation off the machine) with the control group (successful extubation off the machine) (P<0.05); Logistic regression equation showed that age, DTF, DE, E/A, PaO2, PaCO2, CRP/ALB, and basic history of cardiopulmonary disease were all influencing factors for the failure of offline extubation of severe pneumonia (P<0.05). The nomogram prediction model for offline extubation failure for severe pneumonia was obtained by visualization using R language software. The AUC of the nomogram prediction model was 0.866 (95%CI:0.801-0.930) in the training set and 0.917 (95%CI:0.853-0.982) in the verification set, respectively. The calibration curve was close to the 48° reference line, the prediction points were evenly distributed, and the DCA curve was within the range of 0.35 ~ 0.8. The nomogram prediction model could obtain the greatest benefits in both the training set and the verification set.Conclusion: This nomogram prediction model based on diaphragm ultrasound, heart parameters, and blood gas indexes can be used to predict the risk of offline extubation of severe pneumonia at an early stage. Accordingly, appropriate intervention plans can be made and the prognosis is improved.

1525 Relationship between Serum COX-2 PTTG1 TLR4 Levels and Clinicopathological Features Prognosis and Survival of Primary Laryngeal Carcinoma

Objective: To investigate the relationship between serum cyclooxygenase-2 (COX-2), pituitary tumor transforming gene 1 (PTTG1), toll-like receptor 4 (TLR4) levels and clinicopathological features, prognosis, and survival of primary laryngeal cancer.Methods: A total of 105 patients with primary laryngeal cancer and 105 patients with benign laryngeal lesions in Dazhou City Central Hospital of Sichuan Province from May 2021 to March 2023 were selected as observation and control groups, respectively. The serum COX-2, PTTG1 and TLR4 levels of the two groups were compared, and the relationship between serum COX-2, PTTG1 and TLR4 levels, and clinicopathological features was analyzed using multiple linear regression, and the prognostic survival rate of the patients with different serum COX-2, PTTG1, and TLR4 levels was compared in the follow-up of 3 years.Results: The levels of COX-2, PTTG1, and TLR4 in the observation group were (38.89±12.14) ng/L, (140.17±30.25) pg/mL, and (13.94±3.25) ng/mL, respectively, which were higher than those in the control group (17.63±5.09) ng/L, (98.49±15.63) pg/mL, and (8.72±2.13) ng/mL (t=16.549, 12.543, 13.765, all P<0.05). Multiple linear regression analysis showed that clinical stage (partial regression coefficient:0.714, 0.722, 0.719), tumor differentiation degree (partial regression coefficient:0.756, 0.749, 0.798), and lymph node metastasis (partial regression coefficient:0.802, 0.798, 0.713) were the influencing factors for the levels of serum COX-2, PTTG1, and TLR4 in patients with primary laryngeal cancer (P<0.05). The 3-year overall survival rate and overall survival rate of the COX-2 high-level subgroup were 80.77% and 71.15%, respectively, which were significantly higher than those of the low-level subgroup 95.83% and 87.50% (P<0.05); the 3-year overall survival rate and overall survival rate of the PTTG1 high-level subgroup were 80.39% and 68.63%, respectively, which were significantly higher than those of the low-level subgroup 95.92% and 89.80% ( P<0.05); 3-year overall survival rate and overall survival rate of TLR4 high-level subgroup were 78.85% and 71.15%, respectively, which were significantly higher than those of 97.92% and 87.50% in the low-level subgroup (P<0.05).Conclusion: The levels of serum COX-2, PTTG1, and TLR4 are high in patients with primary laryngeal cancer, and their high levels are related to clinical stage, tumor differentiation, and lymph node metastasis. Detection of COX-2, PTTG1, and TLR4 levels can reflect the prognosis and survival of patients to a certain extent.

1530 Assessment of the Value of Laboratory Test Indicators for the Early Diagnosis of Neonatal Sepsis

Objective: To analyze the clinical value of laboratory test indicators in the early diagnosis of neonatal sepsis (NS).Methods: Ninety-one patients with NS and 79 healthy infants treated in our hospital from September 2020 to October 2023 were enrolled in this research. The infants were divided into the control group (n=79) and sepsis group (n=91). The detection results of blood indexes were compared between the two groups. Multivariate logistic regression was used to identify the independent risk factors of NS. The receiver operating characteristic (ROC) curve was plotted to analyze the diagnostic value of laboratory indexes on NS.Results: The total number of white blood cells (WBC), neutrophilicgranulocyte (Neu), neutrophil-lymphocyte ratio (NLR), monocyte-lymphocyte ratio (MLR), procalcitonin (PCT), and C-reactive protein (CRP) in sepsis group were higher than those in control group (P<0.05), while the total number of lymphocyte (Lym) and lymphocyte-leukocyte ratio (LLR) were lower than those in control group (P<0.05), but there was no statistical difference in the total number of monocyte (Mono) between the two groups (P>0.05). Multivariate logistic regression analysis showed that WBC, Neu, Lym, PCT, and CRP were independent risk factors for neonatal NS (P<0.05). ROC curve analysis found that the area under the curve (AUC) of the combination of WBC, Neu, Lym, PCT, and CRP in predicting the occurrence of NS was the highest at 0.960, higher than that detected by the above indicators alone.Conclusion: WBC, Neu, Lym, PCT, and CRP are independent risk factors for neonatal NS, and combined detection has the highest efficiency in the early diagnosis of NS.

1534 Correlation between Serum Mir-647 Levels and In-Stent Restenosis after Bare Metal Stent Placement in Patients with Lower Limb Atherosclerotic Occlusive Disease

Objective: To explore and analyze the association between serum short non-coding RNA-647 (miR-647) level and in-stent restenosis (ISR) in patients with lower extremity arteriosclerosis obliterans (ASO) after bare metal stent implantation.Methods: Totally 182 patients with lower extremity ASO in the hospital were enrolled from September 2020 to September 2023. Serum miR-647 level after bare metal stent implantation was detected, and the correlation between its level change and ISR was analyzed.Results: Out of 182 ASO patients, 43 experienced ISR after metal bare stent implantation, with an incidence rate of 23.63%. The levels of miR-647, neutrophil to lymphocyte ratio (NLR), and C-reactive protein (CRP) in ISR patients were significantly higher than those in non-ISR patients, with statistical significance (P<0.05). Univariate analysis showed statistical differences in Fontaine staging, age, hypertension, and diabetes mellitus between ASO patients with postoperative ISR and patients without ISR (P<0.05). Person correlation analysis showed that miR-647 was positively correlated with NLR and CRP levels (r=0.351, 0.402, P<0.05). Logistic regression analysis showed that Fontaine staging, age, miR-647, NLR, CRP level, hypertension, and diabetes mellitus were risk factors for postoperative ISR in ASO patients (P<0.05). Concerning the ROC curve, the area under the curve (AUC), optimal cut-off value, sensitivity, and specificity of miR-647 expression level in evaluating postoperative ISR were 0.767 (95%CI:0.676-0.858), 0.980, 0.581 and 0.899 respectively.Conclusion: Serum miR-647 expression in patients with ASO after bare metal stent implantation is related to the occurrence of ISR, and has certain predictive and evaluated value on the occurrence of ISR.

1539 Diagnostic Value of High-Frequency Ultrasound Combined with Serum LAG-3 and FGFR-4 in Thyroid Cancer

Objective: To investigate the expression of serum lymphocyte activation gene 3 (LAG-3) and fibroblast growth factor receptor 4 (FGFR-4) in thyroid cancer (TC), and the diagnostic value of them combined with high-frequency ultrasound for TC.Methods: A total of 203 patients with thyroid lesions admitted to our hospital from December 2020 to December 2023 were enrolled as the study subjects. Based on the pathological examination results, they were separated into TC patients as the TC group (n=108) and benign thyroid lesions patients as the non-TC group (n=95). ELISA method was applied to determine the expression levels of serum LAG-3 and FGFR-4. ROC curve was plotted to analyze the diagnostic efficacy of serum LAG-3 and FGFR-4 for TC. Four grid table method was applied to analyze the diagnostic efficacy of high-frequency ultrasound combined with serum LAG-3 and FGFR-4 levels for TC.Results: Compared with undifferentiated thyroid carcinoma, the serum LAG-3 level of papillary, follicular, and medullary thyroid carcinoma was significantly increased, and the serum FGFR-4 level was significantly decreased (P<0.05). Compared with the non-TC group, the serum LAG-3 level in the TC group was reduced, and the serum FGFR-4 level was increased (P<0.05). The levels of PSV and RI in the TC group were higher than those in the non-TC group (P<0.05). The accuracy, sensitivity, and specificity of high-frequency ultrasound combined with serum LAG-3 and FGFR-4 in diagnosing TC were 86.70%, 96.30%, and 75.79%, respectively. Among them, the sensitivity of the three combined diagnoses was higher than that of high-frequency ultrasound, serum LAG-3, and FGFR-4 individual diagnoses (P<0.05).Conclusion: The serum LAG-3 level in TC patients is reduced, while the serum FGFR-4 level is increased. High-frequency ultrasound combined with serum LAG-3 and FGFR-4 has higher diagnostic efficacy for TC.

1545 Changes and Clinical Significance of CTC Level in Patients with Thyroid Cancer before and after 131I Treatment

Objective: To investigate the changes in peripheral blood circulating tumor cell (CTC) levels in patients with thyroid cancer before and after iodine radioisotope (131I) treatment and to analyze their clinical significance.Methods: A total of 108 patients with thyroid cancer who were treated with 131I in the hospital from January to December 2022 were selected as the study subjects. According to the prognosis after 12 months of treatment, the patients were divided into a metastasis group (19 cases) and a non-metastasis group (89 cases). CTC levels before and after treatment and general information of the two groups were comparatively analyzed. Logistic regression analysis was conducted to screen the prognostic factors.Results: CTC levels before and after treatment were (15.51±1.67) FR/mL and (8.50±2.30) FR/mL, with a statistically significant difference (P<0.05). The difference in CTC level in the non-metastasis group before and after treatment was significantly higher than in the metastasis group (P<0.05). There was no statistically significant difference between the two groups in terms of age, gender, length of hospital stay, tumor type, tumor location, capsule invasion, T stage, residual thyroid mass, time from surgery to 131I treatment, frequency of 131I treatment, positive lymph nodes in the lateral cervical region, and Hashimoto's thyroiditis (P>0.05). The difference in the number of positive lymph nodes in the central cervical region was statistically significant (P<0.05). Logistic regression analysis found that the number of positive lymph nodes in the central cervical region was an independent influencing factor for the recurrence and metastasis of thyroid cancer after 131I treatment.Conclusion: 131I treatment after thyroidectomy can effectively improve CTC levels in patients with thyroid cancer, and the improvement of CTC levels before and after treatment is lower in patients with recurrence and metastasis. CTC helps predict recurrence and metastasis after thyroidectomy combined with 131I treatment. The number of positive lymph nodes in the central cervical region is an independent influencing factor for the recurrence and metastasis of thyroid cancer after 131I treatment.

Objective: To observe the changes of oxidative stress indexes and matrix metalloproteinases (MMPs) levels in synovial fluid of patients with ankle trauma, and to analyze their association with pain degree and foot-ankle function.Methods: A total of 163 patients with ankle trauma admitted from February 2020 to February 2023 were enrolled. The levels of oxidative stress indexes [superoxide dismutase (SOD), malondialdehyde (MDA)], MMP-3 and MMP-13 in synovial fluid were measured, and the relationship between their levels and pain degree and foot-ankle function was analyzed.Results: There were statistical differences in the levels of SOD, MDA, MMP-3 and MMP-13 in synovial fluid among patients with different pain degrees (P<0.05). The level of SOD in the pain-free group was higher than that in the mild group and the moderate group (P<0.05) while the levels of MDA, MMP-3 and MMP-13 were significantly lower than those in the mild and moderate groups (P<0.05). The level of SOD in the mild group was higher than that in the moderate group (P<0.05), while the levels of MDA, MMP-3 and MMP-13 were significantly lower than those in the moderate group (P<0.05). The levels of SOD, MDA, MMP-3 and MMP-13 in the synovial fluid of patients with different ankle functions were statistically different (P<0.05). The level of SOD in the excellent group was higher than that in the good group, the fair group and the poor group (P<0.05), while the levels of MDA, MMP-3 and MMP-13 were significantly lower than those in the good group, the fair group and the poor group (P<0.05). The level of SOD in the good group was higher (P<0.05) ,while the levels of MDA, MMP-3 and MMP-13 were significantly lower than those in the fair group and the poor group (P<0.05). The level of SOD in the fair group was higher than that in the poor group (P<0.05) while the levels of MDA, MMP-3 and MMP-13 were significantly lower (P<0.05). Spearman correlation analysis showed that SOD was negatively correlated with ankle pain degree (r=-0.546, P<0.05), and MDA, MMP-3 and MMP-13 were positively correlated with degree of ankle pain (r=0.559, 0.595, 0.517, P<0.05). SOD was positively correlated with ankle function (r=0.495, P<0.05). MDA, MMP-3 and MMP-13 were negatively correlated with ankle function (r=-0.571, -0.574, -0.532, P<0.05). ROC curve revealed that the AUC of the combination of the four indexes of SOD, MDA, MMP-3 and MMP-13 in evaluating the excellent and good rate of ankle function was 0.955, which was significantly higher than the AUC of each single index (P<0.05), and the sensitivity and specificity were 0.835 and 0.944 respectively. Conclusion: There are differences in the levels of SOD, MDA, MMP-3 and MMP-13 in the synovial fluid of ankle trauma patients with different pain degrees and ankle functions, and the above indexes are meaningful to effectively evaluate the recovery of foot-ankle function.

1555 A Study on the Predictive Value of Preoperative Leukocyte Derivativeness Index for Early Recurrence after Hepatocellular Carcinoma Intervention

Objective: To analyze the predictive value of preoperative leukocyte-derived indicators on early recurrence of liver cancer after interventional surgery.Methods: Totally 216 patients who underwent interventional surgery for liver cancer in the hospital were included in the study from January 2020 to January 2023. They were followed up for 1 year after surgery and were divided into a recurrence group (n=110) and a non-recurrence group (n=106) according to the recurrence status. Leukocyte-derived indicators of all study subjects were measured before surgery, including neutrophil-to-lymphocyte ratio (NLR), platelet-to-lymphocyte ratio (PLR), and lymphocyte-to-monocyte ratio (LMR). Subject operating characteristic curve (ROC) was performed to analyze the predictive value of preoperative levels of NLR, PLR, and LMR on early recurrence after interventional surgery for liver cancer.Results: The preoperative NLR and PLR levels in the recurrence group were higher than those in the non-recurrence group while the LMR level was lower than in the non-recurrence group (P<0.05). ROC curve analysis results showed that the area under the curve (AUC) of a combination of preoperative NLR, PLR, and LMR in predicting early recurrence after liver cancer interventional surgery was 0.958, with a sensitivity of 82.73% and a specificity of 95.28%. The efficiency of combined prediction was better than that of each indicator (P<0.05). Conclusion: Preoperative leukocyte-derived indicators are closely related to early recurrence of liver cancer after interventional surgery. Preoperative measurement of NLR, PLR, and LMR levels can predict early recurrence after liver cancer interventional surgery, and provide a certain reference basis for guiding clinical treatment.

1560 Analysis of Factors Influencing the Occurrence of Contrast Reflux during RT-3D-HyCoSy in 125 Cases of Infertility

Objective: To analyze the influencing factors of contrast agent reflux in infertile women during transvaginal real-time three-dimensional hysterosalpingo-contrast sonography (RT-3D-HyCoSy), and to provide a reference for the prevention and treatment of contrast agent reflux.Methods: The case data of 125 patients of infertility who received RT-3D-HyCoSy examination in the hospital were collected from January 2022 to September 2023. According to whether contrast agent reflux occurred, they were divided into the contrast agent reflux group (n=21) and the contrast agent non-reflux group (n=104). Univariate analysis and multivariate Logistic regression analysis were used to analyze the risk factors of contrast agent reflux during RT-3D-HyCoSy.Results: The proportions of patients with secondary infertility, history of intrauterine surgery, adenomyosis, history of ectopic pregnancy, adnexal mass, uterine deformity, moderate-to-severe contrast agent injection pressure, endometrial thickness≤3.5 mm, tube obstruction, and angiography time ≤5 days after menstruation were higher in contrast agent reflux group than those in contrast agent non-reflux group (all P<0.05). Secondary infertility (OR=2.088, 95%CI=1.119~3.893, P=0.021), history of ectopic pregnancy (OR=2.061, 95%CI=1.136~3.739, P=0.018), endometrial thickness≤3.5 mm (OR=2.319, 95%CI=1.186~4.533, P=0.014) and tube obstruction (OR=2.109, 95%CI=1.259~3.531, P=0.005) were the risk factors of contrast agent reflux in infertility women during RT-3D-HyCoSy (P<0.05).Conclusion: Secondary infertility, history of ectopic pregnancy, endometrial thickness≤3.5 mm, and tube obstruction are risk factors of contrast agent reflux in women with infertility during RT-3D-HyCoSy examination. It is necessary to pay clinical attention to the prevention of contrast agent reflux during RT-3D-HyCoSy examination in infertile women using the above risk factors.

1567 Effectiveness of Roxithromycin Combined with Mometasone Furoate Nasal Spray in the Treatment of Bronchial Asthma and Its Effect on Inflammatory Factors and Immune Functions

Objective: To explore the effects of roxithromycin combined with mometasone furoate nasal spray on serum inflammatory factors and immune function in children with bronchial asthma.Methods: A total of 120 children with bronchial asthma diagnosed in the hospital from June 2020 to June 2023 were completely randomized by random number table method into control group and experimental group, the control group was treated with mometasone furoate nasal spray, and the experimental group was treated with roxithromycin combined with mometasone furoate nasal spray for 4 weeks. Lung function, T-cell subpopulation levels, serum inflammatory factors, asthma control test (ACT) scores, quality of life, and adverse effects were compared between the two groups.Results: After treatment, peak expiratory flow (PEF), forced expiratory volume in 1 second (FEV1), and FEV1/forced vital capacity (FVC) were increased in both groups, and changes in amplitude of the above indexes were greater in the experimental group (P<0.05). After treatment, CD3+, CD4+ and CD4+/CD8+ were increased, while CD8+ was decreased in both groups, and changes amplitude of the above indexes was greater in experimental group (P<0.05). After treatment, levels of serum interleukin (IL)-8, IL-13, and tumor necrosis factor-α (TNF-α) were decreased, while levels of serum γ-interferon (IFN-γ), scores of ACT and quality of life were increased in both groups and changes the amplitude of the above indexes was greater in the experimental group (P<0.05). There was no significant difference in the incidence of adverse reactions between the two groups (P>0.05).Conclusion: Roxithromycin combined with mometasone furoate nasal spray can improve pulmonary function, asthma control, and immune function, regulate the inflammatory response, and improve quality of life in children with bronchial asthma, which has good safety.

Objective: To investigate the efficacy of insulin aspart combined with insulin detemir in the treatment of GDM and the effect on serum neuregulin 4 (NRG4), HGF, and amyloid A (SAA) levels and islet cell function.Methods: From June 2020 to June 2023, 113 patients with GDM were selected and randomly divided into the control group (n=56) and the observation group (n=57). The control group was treated with insulin aspart combined with premixed protamine recombinant human insulin while the observation group was treated with insulin aspart combined with insulin detemir. The two groups were compared on insulin use, time to blood glucose reaching the standard, blood glucose indicators (FBG, 2hPBG, and HbA1c), serum NRG4, HGF and SAA levels, islet cell function indicators [HOMA-IR, Homeostasis Model Assessment of beta-cell (HOMA-β) and fasting insulin (FINS)] and pregnancy outcomes. Results: The dosage of insulin and injection frequency in the observation group were lower than in the control group, and the time to blood glucose reaching the standard was shorter than that in the control group (P<0.05). The differences in FBG, 2hPBG, HbA1c, serum NRG4, HGF and SAA levels, HOMA-β, HOMA-IR, and FINS before and after treatment in the observation group were greater than those in the control group (all P<0.05). There was no statistically significant difference in the incidence rates of cesarean section, fetal macrosomia, neonatal hypoglycemia, and fetal distress between the two groups (P>0.05). Conclusion: The combined treatment with insulin aspart and insulin detemir has a significant therapeutic effect on GDM, which can control blood glucose, shorten the time for blood glucose to reach the standard, and improve serum NRG4, HGF, and SAA levels as well as islet cell function.
